The Size Determination of DNA Restriction Fragments kit, 4313B15, demonstrates the electrophoretic techniques for the estimation of DNA fragment sizes, which are essential in the mapping of restriction enzyme sites. This experiment introduces the principles of DNA fragment size determination or measurement. It includes standard DNA markers and fragments of unknown size. The PCR - Polymerase Chain Reaction kit, 4313B23, introduces students to the principles, practice and applications of Polymerase Chain Reaction (PCR). This simulation experiment does not contain human DNA and does not require a thermal cycler. Kit contains 1 tube practice gel loading solution, 1 bottle of Ultra Spec-Agarose Powder, 1 bottle of 50x concentrated electrophoresis buffer, 1 bottle of concentrated Methylene Blue Plus Stain, 1 mL pipet, 100 mL graduated cylinder (packaging for samples) and 10 micro-tipped transfer pipets. All kits require: Horizontal gel electrophoresis apparatus, DC power supply, staining tray and net, visualization system (white light), automatic micropipettes with tips to obtain samples for 6 gels, pipet pump, 250 mL flask or beakers, hot gloves, distilled or deionized water, microwave, hotplate or Bunsen burner, photo-documentation system (optional) and a balance. Kits 4313B01 and B15 also require a water bath.
